Vital role played by fertility 'master gene' found by British scientists

They think this gene helps the fertilised egg decide which cells will go on to form a fetus, and which will form parts of the placenta and yolk sac.

Pioneering British scientists have identified a key gene that may help to explain why so many IVF pregnancies fail.

The study was done under a research licence and strict regulatory oversight from the Human Fertilisation and Embryology Authority (HFEA), the UK Government's independent regulator overseeing infertility treatment and research.

Dr Kathy Niakan, who led the work published in the journal Nature, said: "Our research is the first time that genome editing has been used to understand the role of a gene in early embryonic development". "Pluripotent" stem cells, which can be derived from human embryos, can form any type of tissue in the body and scientists use stem cell technology to create tissue that can fix damage or replace missing structures in the body.

For the first time, a team of scientists used the CRISPR/Cas9 gene editing to study the human embryo and its evolution and development.

The study team used CRISPR/Cas9 to "turn off" the production of one particular gene, known as the OCT4.

But, as the researchers report in Nature, the blastocyst is incapable of fully forming without OCT4, and essentially implodes. After seven days, embryo development was stopped and the embryos were analyzed.

The research was conducted by the Francis Crick Institute, who applied for permission to begin gene editing back in September 2015. In the current study, for example, it's clear that Oct4 is an important gene for helping human embryos develop normally.

Using CRISPR-Cas9, the researchers "turned off" the gene that produces OCT4 and observed what happened to 41 embryo development over the next seven days. Dr Niakan and colleagues are providing new understanding of the genes responsible for a crucial change when groups of cells in the very early embryo first become organised and set on different paths of development. An embryo is only able to implant in the womb after successfully reaching the blastocyst stage, which has three cell types created to develop into the embryo, the placenta, and the yolk sac.

After showing gene-editing techniques can highlight the functionality of certain genes in this way, the team hopes other scientists will use the methods to discover the role of other genes and ultimately help improve IVF and avoid miscarriage.

"If we knew the key genes that embryos need to develop successfully, we could improve IVF treatments and understand some causes of pregnancy failure", said Dr Kathy Niakan from the Francis Crick Institute, who led the research, "It may take many years to achieve such an understanding; our study is just the first step". Many in vitro fertilization efforts also fail at this stage, so it is a critical juncture for researchers to understand.

Those advances are still many years away, says Niakan.

It is illegal in the United Kingdom to edit human embryos for anything other than scientific, ethically-reviewed research.

Niakan calls her team's work simple research.

"This proof-of-principle paper uses CRISPR genome editing to show that, although genetic expression in the early mouse embryo may be similar to a human embryo".
